COURSE DESRIPTION

502-4 (2,2) Research Scanning Electron Microscopy. (a) Theory of design for scanning electron microscope, lenses, vacuum systems, alignment, specimen preparation for biologists and materials scientists, darkroom. (b) Laboratory practical experience in use of scanning electron microscope and specimen preparation. Laboratory fee $100.

COURSE OBJECTIVES

SCI502 or PLB 502, Research Scanning Electron Microscopy, is a graduate level course in the use of the scanning electron microscope as a research tool. It is an introductory course designed to provide sufficient background information to thoughtfully design research protocols, prepare specimens, use instrumentation for the optimal recording of images, and prepare the images for presentation at a professional scientific meeting. The course is divided into 502A (lecture) and 502B (lab). Both may be taken concomitantly or separately if student has appropriate academic or practical experience. Upon satisfactory completion of the introductory course, new users of the facility will receive more, in-depth instruction on instrument use and specimen preparation suited to your specific research project.

LABORATORY FEE

The laboratory fee ($150.00) is due by the fourth week of the course. If this poses problems, divided payments may be made throughout the course until the total amount is paid before the end of the course. This fee covers expenses related to the operation and maintenance of the microscope only.

COURSE INFORMATION

  1. The purpose of the lab course is to train individuals to use the electron microscope in a research project at SIU. The student must have a research project upon signing up for the course.
  2. The student is expected to produce a high quality poster and to give a 15 min presentation of the research for the final lab exam. The poster becomes the property of the IMAGE for several semesters (to display and demo to other students and faculty). 50% of the student's grade will be derived from the poster and the presentation.
  3. Being a graduate level course, the student is expected not only to become proficient in the basic use of the SEM and ancillary equipment but also to understand the operational principles of the microscope and all ancillary equipment. This will necessitate doing outside readings, answering questions and taking quizzes related to principles. In other words, this is not simply a “how to” course, but requires understanding.
  4. The overall product of any electron microscope is an electron micrograph made from a digital file. We will train you how to do this prior to the poster presentation.
  5. Professional electron microscopists are by necessity neat, fastidious and highly-organized individuals. We expect all students to keep their work areas likewise. The appearance of the poster (as well as all micrographs and lab reports) must be well-organized, neat and professional in appearance.
  6. Any reports will be submitted as typewritten documents, and will be due when indicated by the instructor. Quizzes may consist of oral and possibly written questions related to the present or past labs.
  7. Images for the laboratory reports will be produced digitally or on a thermal printer via an SEM image capture system. Images for the poster will be generated digitally. Students will receive two specimen stubs and a stub box.
  8. 50% of grade = laboratory reports. You will be permitted to resubmit one lab a second time to improve your grade.
  9. The recommended textbook for the course is Bozzola and Russell. Other acceptable books covering the topics discussed in the course are also listed below. These may be obtained from Morris Library or used in the library at the IMAGE Center.
