COURSE DESRIPTION
501-4 (2,2) Research Transmission Electron Microscopy. (a) Theory of
design of electron microscope, lenses, vacuum systems, alignment,
specimen preparation and darkroom. (b) Practical experience in use of
transmission electron microscope and specimen preparation.

COURSE OBJECTIVES
SCI or PLB 501, Transmission Electron Microscopy, is a graduate level,
introductory course in theory and practical use of the transmission
electron microscope as a research tool. It is designed to provide suffi
cient background information to: thoughtfully design research
protocols, prepare specimens, use instrumentation for the optimal
recording of images, and prepare the images for presentation at a
professional scientifi c meeting. It is designed for the scientist who
has a SERIOUS need for TEM in their work. After taking this course,
researchers normally continue using the TEM in their research,
eventually becoming independent research users of the TEM at
substantially reduced user fees. In order to become independent user,
it is necessary to take a qualifying examination (written and
practical).
COURSE ORGANIZATION
Each student will be given 2 hours each week of microscope operating
instructions on the Hitachi H7000 TEM in small group of 3-4 trainees.
In addition, regular meetings involving the entire class will be held
every week in order to assess student progress in obtaining
and preparing a specimen as well as provide input from all students
concerning problems encountered and potential solutions. Individualized
instruction in specifi c techniques will be available and is the
responsibility of the student to arrange. Please be aware that certain
specialized techniques (ion beam or mechanical thinning,
cryo-microscopy, etc.) are beyond our capabilities due to lack of
equipment. Specialized equipment may, however, be available in various
departments on campus.
LABORATORY FEE
We have a laboratory fee of $150.00 per trainee, to partially defray
cost of microscope usage. The actual cost of instruction is well over
$1,500. The fee must be paid by the third week of the course. It may be
paid by personal check made out to SIU or, if desired, it may be paid
from a university account. We will prepare invoice vouchers so that
faculty sponsors with grants or university accounts may pay for the
course.
